摘要
目的通过观察白细胞介素-1(interleukin-1,IL-1)对房水流出通道中基质金属蛋白酶(matrix metalloproteinases,MMPs)的影响,从分子细胞学水平探讨IL-1的降眼压机制。方法SD大鼠10只,随机选择一眼前房注射重组鼠IL-1β30ng,对侧眼前房注射等体积1g·L-1PBS作为对照。用药30h后取眼球,进行免疫组织化学检测房水流出通道的MMP-9表达,并利用自动图像分析系统对实验结果进行半定量分析。结果用药眼的小梁网、睫状肌和虹膜均出现强阳性反应,自动图像分析显示用药眼与对侧眼的灰度分别为47.39±3.30、88.42±4.76,差别有非常显著统计学意义(t=25.95,P=0.000<0.01)。结论IL-1降眼压机制可能与3条房水流出通道的MMPs表达增加有关。
Objective To determine the underlying cellular and molecular mechanisms of interleukin-1 (IL-1) on intraocular pressure reduction by studying its effects on matrix metalloproteinases (MMPs) in aqueous humor outflow pathways.Methods Ten Sprague-Dawley rats were selected with one eye receiving intracamerally injection of 30 ng IL-1β and the control ateral eye receiving equivalent volume of 1 g·L^-1 PBS intracamerally injection.Eyes were removed 30 hours after treatment,then immunohistochemistry was done to examine the expression of MMP-9 in aqueous outflow pathway,and the experimental results were semiquantitative analyzed with computerized image analysis techniques.Results Strong positive reaction were detected in trabecular meshwork,cilliary muscle and iris of the treated eyes.The gray scale resolutions of the treated eyes and the contralateral eyes were 47.39±3.30 and 88.42±4.76,respectively,which was statistically different (t=25.95,P=0.000〈0.01).Conclusion Increased expression of MMPs in three aqueous humor outflow pathways may be the underlying IOP reduction mechanisms of IL-1.
