金雀黄素诱导人子宫内膜癌细胞HEC-1B凋亡及机制的研究 被引量:2

摘要 目的研究金雀黄素(Genistein,GEN)诱导人子宫内膜癌细胞株HEC-1B凋亡,探讨金雀黄素诱导HEC-1B细胞凋亡与抑癌基因PTEN之间的关系。方法应用流式细胞仪检测细胞凋亡,应用流式细胞仪检测抑癌基因PTEN蛋白的表达,两者之间的相关性。结果金雀黄素诱导HEC-1B细胞的凋亡,其作用随剂量增加而增强;金雀黄素可上调HEC-1B细胞抑癌基因PTEN蛋白的表达,其作用随剂量增加而增强。结论金雀黄素能够诱导HEC-1B细胞凋亡,其机制可能是通过上调HEC-1B细胞PTEN的表达而实现。 Objective To investigate the effect of Genistein on human endometrial carcinoma cell line HEC - 1B and its relationship with expression of the anti - oncogene PTEN, Methods The apoptosis cells and expression of PTEN protein were determined by flow cytometry. Results Genistein induced apoptosis of HEC- 1B in a dose - dependent manner. It also increased the expression of PTEN protein in a dose - dependent manner. Conclusions Genisteln can induce the apoptosis of HEC - 1B cell, which might be mediated by up - regulation of anti - gene PTEN.
