摘要
目的进一步明确天然糖皮质激素对巨噬细胞免疫功能的影响及其时效和量效关系。方法收集大鼠腹腔巨噬细胞,分别用10~1000ng/ml的皮质酮(corticosterone,CORT)刺激,检测细胞的黏附、趋化和吞噬功能;使用ELISA试剂盒检测细胞培养上清中TNF-α的含量。结果50ng/ml的皮质酮显著增强巨噬细胞的黏附功能(P〈0.05),10ng/ml和50ng/ml的皮质酮刺激细胞后,其趋化功能均显著增加(P〈0.05)。同时,50ng/ml的皮质酮还可显著增强巨噬细胞的吞噬功能(P〈0.05)和促进巨噬细胞分泌TNF-α(P〈0.01),而使用高浓度皮质酮(500ng/ml和1000ng/m1)处理时,巨噬细胞的上述功能增强趋势减弱或无增强;在0.5—5h范围内,皮质酮处理1h时细胞的吞噬功能和分泌TNF-α的功能增强最明显,刺激更长时间后其功能的增强趋势逐渐减弱。结论一定剂量的天然糖皮质激素急性刺激可明显增强免疫功能。
Objective To investigate the effect of endogenous glucocorticoids on the immune function of macrophages and their dose-effect or time-effect relationship. Methods The peritoneal macrophages obtained from male SD rats were stimulated with corticosterone ( 10, 50, 100,500, 1000 ng/ml) for different time period. Then the innate immunity of macrophages was evaluated, such as adherence, Chemotaxis, phagocytic capacity and the ability to secret TNF-α. Results Corticosterone (50 ng/ml) enhanced adherence and phagocytic ability of macrophages (P 〈 0.05 ) and their ability to secret TNF-α (P 〈 0. 01 ). Corticosterone ( l0 ng/ ml and 50 ng/ml) enhanced Chemotaxis of macrophages (P 〈0. 05). When stimulated by 500 or 1000 ng/ml corticosterone, the immune function of macrophages was depressed or not influenced. Within the time period of stimulation from 0.5 to 5 h, the corticostetone treatment for 1 h could improve the phagocytic function and TNF-α secretion of peritoneal maerophages, but longer stimulation suppressed this effect. Conclusion Acute stimulation of endogenous glucocorticoids at a certain dosage can enhance the immune function.
