摘要 目的:探讨氰基丙烯酸正辛酯/正丁酯(N-OCA/N-BCA)在眼组织的生物相容性。方法:仿青光眼小梁切除术制作兔模型,将N-OCA/N-BCA涂抹于小梁和虹膜切除后的角巩膜伤口边缘和巩膜层间,展平巩膜瓣到剖切床上使其牢固粘合,术后行裂隙灯观察、组织病理学、超微结构和TUNEL法检测虹膜睫状体和视网膜细胞凋亡,分别用单纯剖切和缝线做为对照组。结果:术后裂隙灯观察各组早期结膜均轻度充血水肿、分泌物少、前房未见明显反应。术后各个时间段N-OCA/N-BCA组兔眼组织病理学和超微结构变化、对角膜内皮细胞的毒性和TUNEL法检测细胞凋亡等方面于单纯剖切组相比差异无显著性(P>0.05);巩膜瓣于术后7,14d尚未愈合,缝线组已愈合;术后30d组织学改变巩膜瓣的愈合相似于14d缝线组。结论:N-OCA/N-BCA在兔眼组织内有良好的生物相容性,可以延缓巩膜瓣的愈合。 AIM: To observe and investigate the biocompatibility of the N-Octyl-Cyanoacrylate/N-Butyl-Cyanoacrylate (N-OCA/N-BCA) in ocular tissue. METHODS: The rabbit model was constructed simulating trabeculectomy. N-OCA/N-BCA was squashed between the scleral flap and dissected bed, and the flattening scleral flap was quickly pasted to the dissected bed firmly. After the surgery, we observed the operated eyes by slit-lamp microscope, histopathology, ultrastructure examination and TUNEL assay to detect apoptosis of corpus ciliare choroideae and retina, using simple dissected group and suture line group as control.RESULTS: In the early period after the surgery, slit lamp observation showed that each group had light hyperemia and edema in conjunctiva, little secretion, and no conspicuous reactions in the anterior chamber. Between the N-OCA/N-BCA group and simple dissected group, there were no significant differences (P〉0.05) in histopathology, ultrastructure, corneal endothelial cells toxicity and apoptosis at various time points after the surgery. The scleral flaps of the N-OCA/N-BCA group were not healed at 7 and 14 days after the surgery while they were already healed in suture line group. The healing of scleral flaps which had histological alteration at 30 days after the surgery was similar to that of suture line group at 14 days after the surgery.CONCLUSION: It has been initially proved that N-OCA/ N-BCA had satisfactory biocompatibility in the rabbit ocular tissue, and it could delay the healing of the scleral flap.
