摘要
目的观察重组人酸性成纤维细胞生长因子(rhaFGF)对家兔体表溃疡愈合的促进作用。方法建立家兔的体表溃疡模型,其中2组分别按90,60 AU.(cm2)-1的剂量给予rhaFGF,阳性对照组按60 AU.(cm2)-1的剂量给予重组牛碱性成纤维细胞生长因子(rb-bFGF),阴性对照组给予与第二组等量的溶媒。采用创面照相、透明膜描记称量法记录溃疡形成后第1,4,8天溃疡面面积,比较面积收缩量;用注水法测量伤腔容积,比较容积缩小量。溃疡形成后第8和第14天取创面组织,经10%甲醛溶液固定后制片,常规染色,重点观察创面肉芽组织生长情况,包括毛细血管生长、胶原沉积与再上皮化变化,以评价修复效果。结果各组家兔溃疡面面积随时间的延长而逐渐缩小,其中rhaFGF 60AU.(cm2)-1组和rb-bFGF60 AU.(cm2)-1组疗效更显著(均P<0.05)。各组家兔伤腔容积随伤后时间的延长而逐渐缩小,其中rhaFGF 60 AU.(cm2)-1组和rb-bFGF 60 AU.(cm2)-1组更为明显(均P<0.01)。组织学检查,可见rhaFGF60 AU.(cm2)-1组和rb-bFGF60 AU.(cm2)-1组治疗8 d后家兔的溃疡面成纤维细胞生长活跃,数量多,毛细血管胚芽与成纤维细胞数量显著多于阴性对照组;第14天,rhaFGF60 AU.(cm2)-1组和rb-bFGF60 AU.(cm2)-1组家兔溃疡面创面收缩和再上皮化明显,且优于阴性对照组。结论rhaFGF对家兔体表溃疡有促进愈合作用,可促进创面肉芽组织生长、毛细血管胚芽形成与再上皮化。
Objective To study the effect of recombinant human acidic fibroblast growth factor (rhaFGF) on promoting rehabilitation of skin ulcerations in rabbits. Methods The skin ulceration models of rabbits were made and divided into groups, among which two groups received 90, 60 AU ·(cm^2)^-1 rhaFGF, the third positive control group received 60 AU ·(cm^2)^-1 rb-bFGF, and the negative control group received only the vehicle of the equal quantity as the second group. The ulceration area was recorded on photos and cellophane film at 1,4,8 d, and the volume of injured cavity was measured with water injection. The reduced degree of the area and the volume were compared as well. At 8,14 d, the growth of wound granulation and re-epithelization were tested to evaluate the effects of rhaFGF. Results The skin ulcerations of the rabbits became smaller as time went on in all groups, but 60 AU ·(cm^2)^-1 rhaFGF and 60 AU·(cm^2)^-1 rb-bFGF groups are more significantly than the negative control group. The volume of injured cavity also grew smaller, and 60 AU ·(cm^2)^-1 rhaFGF and 60 AU·(cm^2)^-1 rb- bFGF groups are more significantly. 8 d later, fibroblasts and capillary granulation tissue grew more and more actively, in 60 AU ·(cm^2)^-1 rhaFGF and rb-bFGF groups, than those in the negative group. After 14 d, the ulceration contracted and reepithelized in 60 AU ·(cm^2)^-1 rhaFGF and rb-bFGF groups were more significantly than those in the negative group. Conclusion rhaFGF can promote rehabilitation of the skin ulcerations in rabbits by stimulating the growth of granulation tissue, capillary angiogenesis and of epithelium.
