摘要
目的探讨选择性环氧化酶-2(COX-2)抑制剂尼美舒利(ni mesulide,NI M)对人舌鳞癌Tca8113细胞生存素(Sur-vivin)及血管内皮生长因子(VEGF)表达的影响。方法体外培养人舌鳞癌Tca8113细胞,对照组加入0.4%二甲基亚砜(DMSO),试验组加入不同浓度的NI M(25,50,100,200μmol/L)进行干预,作用24,48,72h后,采用免疫细胞化学法检测舌鳞癌细胞内Survivin及VEGF蛋白表达变化。结果不同浓度的NI M(浓度分别为25,50,100,200μmol/L)处理Tca8113细胞24h,48h,72h后,细胞内Survivin及VEGF蛋白表达均下降。在同浓度下,随着NI M干预时间的延长,Survivin及VEGF的表达较对照组下降更为明显(P<0.05);在同一时间点,随着NI M浓度的增加,Survivin及VEGF表达较对照组亦均明显下降(P<0.05)。经方差分析,二者均呈浓度和时间依赖性。VEGF与Survivin高度相关,相关系数r=0.93(P<0.01)。结论NI M对Tca8113细胞Survivin和VEGF表达的抑制可能是其具有抗肿瘤作用的机制之一。
Objective To investigate the influence of the selective cyclooxygenase-2(COX-2) inhibitor, nimesulide(NIM), on the expression of Survivin and vascular endothelial growth factor(VEGF ) in human tongue squarnous cell carcinoma Tca8113 cell lines. Methods Tca8113 ceils were cultured in vitro with 0.4% Dimethyl Sulfoxide(DMSO) and NIM(25,50,100,200μmol/L) for 24, 48, 72 h in control group and experimental group, respectively. Expression of Survivin and VEGF in Tca8113 cell was determined by immunohistochemistry staining at 24, 48, 72 h. Results At the same concentration, the expression of VEGF and Survivin were significantly down-regulated at different times compared to control group (P 〈 0.05 ). At the same time point, Survivin and the VEGF expression were also significantly down-regulated compared to control group ( P 〈 0.05). The analysis of variance showed that the expression of Survivin and VEGF decreased in a concentration- and time-dependent manner. The variational tenden- cy of VEGF and Survivin was coincident, r = 0.93 (P〈 0.01). Conclusion The inhibitive effect of NIM on the expre.ssion of Survivin and VEGF may be one of important mechanisms for antiumor activity.
