摘要 目的体外培养人胚胎生殖细胞(EGC),通过检测培养的细胞中转录因子Oct-4、端粒酶的表达,鉴定EGC。方法取5~10周人胚胎的生殖腺嵴和肠背系膜,进行组织块体外培养,采用免疫细胞化学检测Oct-4、端粒酶表达。结果组织块体外培养得到的细胞,经免疫细胞化学法显示的单个细胞或细胞集落,均阳性表达转录因子Oct-4、端粒酶。结论组织块体外培养所获得细胞来源于原始生殖细胞(PGCs),为未分化的EGC。 Objective To culture human embryonic germ cells in vitro and identify them by observing the expression of the surface markers Oct-4 and telomerase. Methods The gonadal ridges and mesenteries of human embryos on the 5th to 10th post-fertilization week were isolated and cultured in vitro. Immunocyto6hemistry was applied to detect whether the cells cultured express Oct- 4 and telomerase. Results Both the single cell and the cellular colonies cultured in vitro expressed Oct-4 and telomerase. Conclusion The cells we obtained through tissue culturing are undifferentiated human embryonic germ cells, which are originated from hPGCs.
