摘要
目的探讨抑癌基因PTEN、基质金属蛋白酶9(MMP-9)在脑星形细胞肿瘤中的表达及与肿瘤侵袭性的相关性。方法用免疫组化Elivision法检测PTEN、MMP-9在脑星形细胞肿瘤中的表达情况;同时通过测量核磁共振影像上的瘤周水肿低信号范围,并与免疫组织化学表达结果作比较分析。结果(1)41例人脑脑星形细胞肿瘤标本中,PTEN在瘤细胞的阳性表达率为51.22%,随着肿瘤的恶性程度的增加而降低(P<0.01);(2)MMP-9在瘤细胞的阳性表达率为48.78%,且随着肿瘤恶性程度的增加而增加(P<0.01);(3)在星形细胞肿瘤瘤细胞中的MMP-9表达与肿瘤侵袭性指标MRI扫描图像上表现的瘤周水肿低信号范围分级呈正相关(P<0.05),PTEN表达增加与肿瘤侵袭性指标MRI扫描图像上瘤周水肿低信号范围分级的降低呈负相关(P<0.01)。结论MMP-9在人脑星形细胞肿瘤侵袭中起着一定的作用,PTEN的失活与星形细胞肿瘤的进展关系密切,可以作为脑星形细胞肿瘤的预后判断指标和侵袭指标之一。
Objective To investigate the the relationship of PTEN and matrix metalloproteinase-9 (MMP-9) with the invasion of astrocytic gliomas. Methods Elivision immunohistochemical method was used to measure the expression of PTEN and MMP-9 in 41 astrocytic gliomas in different grades, and in the peritumoral low-density area seen on MRI scan. The relationship of immunohistochemical result and the peritumoral low-density area was analyzed. Results In 41astrocytic gliomas, the expression of PTEN was 51.22% of cases, which was negatively correlated with the grade of astrocytic gliomas(P〈0. 05). The expression of MMP-9 was 48. 78% of cases,which was positively correlated with the grade of astrocytic gliomas(P〈0. 05). In 41 astrocytic gliomas, the expression of MMP-9 was positively correlated with the grade of the peritumoral low-density area(P〈0. 05), and the expression of PTEN was negatively correlated with the grade of the peritumoral low-density area(P%0. 01). Conclusion MMP-9 may paly an important role in the invasion of astrocytic gliomas, and the loss of PTEN is assscoiated with the development of the astrocytic gliomas. Survivin, MMP-9 and PTEN may serve as the indicators of invasion and malignant phenotype.
