摘要
目的:研究肥大细胞膜稳定剂色甘酸钠对缺血再灌注大鼠肠组织组胺、MDA和SOD水平的影响。方法:32只SD大鼠随机分为4组,假手术组(sham组)、模型组(model组)、模型+色甘酸钠50mg/kg组(C1组)及模型+色甘酸钠25mg/kg组(C2组)。复制肠缺血再灌注损伤模型,色甘酸钠于再灌注前15min分别腹腔给予,观察肠黏膜病理结构变化和肠黏膜肥大细胞(IMMC)超微结构变化,测定小肠组织SOD活性、MDA和组胺浓度。结果:(1)model组肠黏膜结构破坏严重,Chiu’s评分明显高于sham组(P<0.01),经色甘酸钠处理组Chiu’s评分低于model组(P<0.01),C1组Chiu’s评分低于C2组(P<0.05)。(2)sham组IMMC超微结构正常,model组IMMC胞浆颗粒明显减少,颗粒包膜相互融合形成细胞内空泡等脱颗粒现象,C1、C2组IMMC超微结构形成空泡较少。(3)model、C2组的小肠组织组胺浓度明显低于sham组(P<0.05);C1及C2组小肠组织组胺浓度明显高于model组(P<0.01)。(4)model组SOD活性最低,sham组最高(P<0.05),C1、C22组间无明显差异(P>0.05);model组的小肠组织MDA浓度明显高于其余3组(P<0.01)。(5)相关分析显示Chiu’s评分、MDA与小肠组胺浓度呈负相关(r=-0.770,P<0.01;r=-0.412,P<0.05),SOD活性与组胺浓度呈正相关(r=0.579,P<0.01)。结论:色甘酸钠能够稳定IMMC,减轻肠缺血再灌注氧化损伤。
AIM: To investigate the effects of mast cell membrane stabilizer on histamine concentration, SOD activity and MDA content in small intestine after ischemia- reperfusion injury in rats. METHODS: 32 SD rats were randomly divided into four groups: sham group; model group; high dosage group (treated with cromolyn sodium 50 mg · kg^-1 , C1 group) ; low dosage group (treated with cromolyn sodium 25 mg · kg^-1, C2 group). The intestinal injury was induced by clamping the superior mesenteric artery for 45 min and reperfusion for 60 min. Cromolyn sodium was administrated via intaperitoneal 15 min before reperfusion. At the end of the study, the intestinal pathology and ultrastructure of IMMC were observed. The levels of MDA, histamine, and SOD activity of the intestine were detected. RESULTS : ( 1 ) Chiu' s score in model group was higher than that in other groups (P 〈 0. 01 ). The high dosage of cromolyn sodium provided better protective effects than the low dosage ( P 〈 0. 05). (2) IMMC in model group was degranulated. Cromolyn sodium attenuated the degranulation. (3) MDA content was significantly increased. SOD activity and concentration of histamine were significantly decreased in model group ( P 〈 0. 05). Cromolyn sodium attenuated the increase in MDA content, decrease in SOD activity and concentration of histamine (P 〈 0.05). (4) A negative correlation between the Chiu' s score and the concentration of histamine ( r = -0. 770, P 〈 0. 01 ), a negative correlation between MDA content and the concentration of histamine (r = -0. 412, P 〈 0. 05 ), and a positive correlation between the SOD activity and the concentration of histamine in the four groups (r =0. 579,P 〈0. 01 ) were observed. CONCLUSION: Cromolyn sodium can inhibit the activity of IMMC, reduce the oxidative products induced by intestinal ischemia - reperfusion.
