人卵巢癌细胞系SKOV3对树突细胞免疫功能及凋亡的影响 被引量:3

摘要 目的:探讨人卵巢癌细胞系SKOV3对树突细胞(DC)免疫表型、细胞因子分泌及凋亡的影响。方法:采用外周血单个核细胞培养DC,以PBS溶液为对照,应用流式细胞仪检测人卵巢癌细胞系SKOV3培养上清液作用于DC后,其凋亡的发生率、免疫表型的表达,应用酶标记免疫吸附分析法(ELISA)检测DC白细胞介素-12(IL-12)的分泌情况。结果:人卵巢癌细胞系SKOV3培养上清液作用于DC后,可诱导DC发生凋亡,DC的CD80、CD83及人类白细胞(位点)DR抗原(HLA-DR)等表型明显下调,IL-12分泌明显下降,与对照组相比差异显著(P<0.05),且IL-12分泌随SKOV3作用时间的延长,其抑制作用更加明显。结论:人卵巢癌细胞系SKOV3可明显抑制DC的免疫功能,并诱导其凋亡。 Objective: To investigate the effects of human ovarian cancer cell line SKOV3 on the immune phenotype, the secretion of cytokine and the apoptosis of dendritic cells (DC). Methods : DC were produced from peripheral blood and observed by flow cytometry for their apoptosis rate and immune phenotype after incubated with the suppernatant of human ovarian cancer cell line SKOV3, with phosphate buffer solution as the control. The level of IL-12 secreted by DC was measured by ELISA. Results: Compared with the control, the apoptosis rate of DC increased after incubated with the supernatants of human ovarian cancer cell line SKOV3, the expressions of HLA-DR, CD80 and CD83 of DC and the level of IL-12 secreted in the tumor environment decreased significantly ( P 〈 0.05 ) , and the inhibitory effect of SKOV3 on IL-12 secretion was more obvious with the prolonging of its action time. Conclusion: Human ovarian cancer cell line SKOV3 can significantly reduce the immune function and induce the apoptosis of dendritic cells.
