金标免疫渗滤法检测猫狗血清中抗肺吸虫IgG

Detection of Anti-paragonimus IgG in Feline or Canine Sera by Dot Immunogold Filtration Assay

目的探索简易、快速的猫、狗肺吸虫病免疫诊断方法。方法以肺吸虫成虫可溶性抗原点样于固相硝酸纤维素膜上,以胶体金-protein A为检测标记物,采用自行设计的渗滤装置,检测猫、狗血清中抗肺吸虫IgG抗体。结果检测7份肺吸虫感染猫血清和6份感染狗血清均为阳性,检测25份健康猫血清和27份健康狗血清均为阴性。结论金标免疫渗滤法快速检测猫、狗血清中肺吸虫特异性IgG,不仅操作简便、快速,结果判读容易,不需特殊仪器设备,而且敏感特异,有潜在的诊断价值,值得进一步开发应用。

Objective To develope a simple and rapid assay for immunodiagnosis of feline or canine paragonimiasis. Methods Soluable adult worm antigen of Paragonimus westermani was dotted on nitrocellulose and protein A coniugated with colloidal gold was used as probe. A filtration device with vertical flow was applied to detect specific IgG against P. westermani in canine or feline sera. Results All of 7 infected feline sera and 6 infected canine sera showed positive. 25 sera from healthy cats and 27 sera from healthy dogs were negative. Conclusion Dot immunogold filtration assay for detection of paragonimus--specific IgG in feline or canine sera was not only simple, rapid, easy to read test results without requirement of any intruement, but also sensitive and specifitive. It would be valuable to be developed and applied widely for diagnosis of canine and feline paragonimiasis.
