雷公藤多甙对白细胞介素-1β诱导大鼠滑膜细胞株RSC-364基质金属蛋白酶-3、c-Jun基因表达的影响 被引量:6

The effect of tripterygium glycosides on the expression of MMP3 and c-Jun mRNA in rat synovial cell line RSC-364 stimulated with IL-1β

摘要 目的 观察雷公藤多甙对白细胞介素(IL)-1β诱导大鼠滑膜细胞株RSC-364基质金属蛋白酶-3(MMP3)、c—Jun mRNA表达的影响。方法 选择不同浓度的雷公藤多甙作用IL-1β刺激的大鼠滑膜细胞株RSC-364,37℃,5%CO2的环境下培养48h,运用酶联免疫吸附试验(ELISA)检测细胞上清的MMP3和实时聚合酶链反应(real—time PCR)检测分析细胞中的c—Jun mRNA表达。结果 不同浓度的雷公藤多甙(0、5、10、20mg/L)对IL-1β刺激的大鼠滑膜细胞株RSC-364分泌MMP3的水平依次是198、176、140、115μg/L,c—Jun mRNA表达分别是1.17×10^6、3.31×10^5、9.32×10^4、2.81×10^4 copies/ml,即呈现明显地抑制。重要的是:c—Jun mRNA表达抑制呈现明显的剂量依赖性。结论雷公藤多甙治疗类风湿性关节炎的有效的作用机制,可能与其抑制MMP3、c—Jun基因的表达相关。 Objective To explore the effect of tripterygium glycosides (TG) on theexpression of matrix metalloproteinase -3 (MMP3) and c-Jun mRNA in rat synovial cell line RSC-364 stimulated with interleukin ( IL)-1β (IL-1β) and to illuminate the molecular mechanism of TG in the treatment of rheumatoid arthritis (RA). Methods The rat synvoial cell line RSC-364 stimulated with IL-1β ( 10μg/L) was induced by different doses of TG (0,5,10 and 20 mg/L) and at 37℃ under 5% CO2 atmosphere was incubated for 48 h. MMP3 in cell supernatant was measured using enzyme-linked immunosorbent assays and the expression of c-Jun mRNA was detected by using real-time reverse transcription polymerase chain reaction. Results The expression of MMP3 in the rat synovial cell line RSC-364 stimulated with IL-1βwas 198,176,140,115 μg/L respectively by different doses of TG (0,5,10 and 20 mg/L),and the expression of e-Jun mRNA was 1. 17× 10^6, 3.31 × 10^5, 9.32×10^4, 2.81 ×10^4 copies/ml respectively, which were suppressed obviously; Of note ,the suppression of the expression of c-Jun mRNA was dose-dependant. Conclusion The effective mechanism of TG in the treatment of RA might be related with the suppression of the expression of MMP3 and c-Jun mRNA.
