云南省肠道病毒96型的分子生物学鉴定 被引量:7

Molecular Identification of Human Enterovirus 96 in Yunnan Province

摘要 目的对云南省9例急性弛缓性麻痹病例的阳性病毒分离物进行分子生物学定型。方法用肠道病毒(En-terovirus,EV)通用引物187、188、189/222扩增病毒VP1区,直接对聚合酶链反应产物进行测序。所得序列在基因库进行广泛搜索,并与所有EV(包括新发现的EV)基因的相同区域进行比较。结果9株病毒为EV96型。结论运用分子生物学方法可以检测出传统方法所无法鉴定的血清型,并能提供病毒遗传学信息。 Objective Molecular typing was conducted according to the reported methods for 9 enteroviruses that were unidentifiable by traditional neutralization method. Methods RT-PCR and sequencing were done according to the Oberste's methods by using 187,188,189/222 primers. Blast searching was done on the Genbank database and the sequences were compared with all available enterovirus database. Results Nine untypeable enteroviruses were proven to be human enterovirus 96. Molecular typing method can not only type the enteroviruses but also provide virus' molecular information.
