栀子总环烯醚萜苷对脑出血大鼠血肿周围NF-κB表达的影响 被引量:11

摘要 目的:研究栀子总环烯醚萜苷对大鼠脑出血后核转录因子κB(NF-κB)表达影响。方法:将SD大鼠随机分为假手术组、脑出血模型组、尼膜同组、栀子总环烯醚萜苷(20、10mg、5mg/kg)治疗组,采用自体血注入诱发大鼠脑出血模型,每组分别在术后12、24、48和96h 4个时间点取脑组织标本,免疫组化检测NF-KB蛋白表达,同时测量脑组织含水量。结果:20~10mg/kg栀子环烯醚萜苷治疗组脑出血后NF-κB P65表达水平及脑含水量明显低于脑出血组。结论:栀子环烯醚萜苷可以抑制脑出血后NF-κB活化,减少炎症因子释放,从而减轻脑水肿。 Objective: To study the influence of total iridoid glycosides in Gardenia jasminoides(TIGGJ) on the expression of NF-κB in perihematomal brain regions of rats. Methods: Male Sprague-Dawley rats were randomly divided into pseudo-operation group, model group, nimotop group and Gardenia jasminoides total iridoid glycosides groups( 20,10 and 5mg/kg). ICH models were induced in rats by injecting stereotactic infusion autologous blood 50μl into pallidus. The brain tissue specments in every group were respectively gathered at the time of 12h ,24h, 48h and 96h after operation. The expression of NF-κB in rat after ICH was detected with inmunohistochemistry method, at the same time the moisture in brain tissue was measured. Resulta: Compared with ICH group,the expression of NF-κB and moisture in brain tissue in (20 , 10mg/kg) were obviously decreased. Conclusion: TIGGT can reduce the expression of NF-κB in rat after ICH, which may be related with the anti-edema effect folloming ICH.
