摘要
目的研究海仁藻酸(KA)致痫老年大鼠海马NMDA受体亚单位1(NMDAR1)蛋白表达的变化,为进一步探明癫痫发病及损伤的机制奠定基础。方法在立体定位仪下,将KA注射至老年大鼠杏仁核制备癫痫模型。将老年大鼠随机分为正常对照组,假手术组及致痫组,分别于不同时间取材,应用流式细胞仪技术及免疫组化的方法观察脑组织NMDAR1蛋白表达的变化。结果正常海马各区均有极少量的NMDAR1阳性细胞分布,KA致痫2 h组老年大鼠海马各区及颞叶皮层NMDAR1表达迅速增加,6 h达高峰,并持续至24 h(P≤0.01)。结论KA致痫后老年大鼠海马NMDAR1蛋白的表达水平上调,并持续至致痫后的24 h,NMDAR1参与了癫痫发生和癫痫脑的长时程的兴奋过程。
Objective By observing the protein expression of N-methyl-D-aspartate receptor (NMDAR) 1 in hippocampus of the elderly epilepsy rats to explore its mechanism. Methods Epilepsy rat models were made by injecting kainic acid into amygdala under stereotactic instrument. 60 male Wistar rats were divided randomly into normal control group, false operation group and epilepsy group. NMDAR1 protein expression were obtained at different time and tested by immunohistochemistry and FAC scan flow cell analysis. Results In the normal group and false operation group, NMDAR1 positive cells distributed rarely in hippocampus ; NMDAR1 positive cells of epilepsy groups were significantly higher 2 h later, reached peak at 6 h, persisting to 24 h (P≤0. 01 ). Conclusions NMDAR1 protein expression is upregulated in epilepsy rats brain and persists for 24 h, which could participate in the excitation process of long time course in brain of the epilepsy rats.
