舒林酸对结肠癌Lovo细胞生长增殖及凋亡的作用研究

Proliferative and proapoptotic effect of sulindac on colonic cancer cell line Lovo

目的研究非甾体抗炎药舒林酸对结肠癌细胞Lovo增殖和凋亡的影响,并探讨其作用机制。方法用不同浓度舒林酸(0.6,0.9,1.2 mmol/L)处理人结肠癌细胞株Lovo,未加药组为对照组,培养不同时间后,采用MTT法测定细胞的生长活性,采用流式细胞仪、透射电镜和吖啶橙染色观察舒林酸对Lovo凋亡作用及周期的影响。结果舒林酸能明显抑制肿瘤细胞生长,使细胞失去正常形态,变小变圆漂浮于培养液中;随舒林酸浓度和作用时间的增加其抑制率增加,与对照组比较有显著性差异(P<0.0 5);透射电镜和吖啶橙染色观察到舒林酸处理后的细胞出现典型的凋亡形态学改变,有凋亡小体形成;FCM测得舒林酸处后Lovo细胞的凋亡率明显增加,且呈时间和剂量依赖性;舒林酸能改变细胞周期分布,使G_0/G_1期细胞增多,S期和G 2/M细胞减少。结论舒林酸具有抑制Lovo细胞增殖的作用,其机制可能与改变细胞周期分布、诱导细胞发生凋亡有关。

Aim：To investigate the effect of sulindac on the proliferation and apoptosis of Lovo cell line and its mechanism. Methods Human colonic cell line Lovo was treated with sulindac （0.6, 0.9, 1.2mmol/L） for various periods in vitro. The growth of Lovo cells tested by MTT assay before and after sulindae treatment respectively. The apoptosis and cells cycle of Lovo cells was analyzed by flow cytometry,electron microscope and acridine orange staining. ResultsLovo cells treated with sulindac displayed a slowed growth in comparison with the control cells, and the inhibitory rate increased accordingly with the increase of sulindac concentration . The apoptotic rate of Lovo cells which were treated with sulindac was increased greatly ,which was markedly higher than that of the controls （1.81±0.91/2.06±1.43） （P〈0.05）. Sulindac significantly increased the proportions of cells at the G0/G1 phase and decreased the proportions of S and G2/M phases and all in a concentration-dependent manner. Conclusion This study has demonstrated that slindac can inhibit the proliferation of Love cells, its anti-tumor mechanism may attribute to change cell cycle,distribution and induce apoptosis .
