摘要
目的:研究Ca2+通道阻滞剂-硝苯地平在正畸牙周膜重建过程中对基质金属蛋白酶(matrix metalloproteinase,MMP)表达与定位的影响。方法:选取60只雄性SD大鼠随机分为加力组、加力+低浓度硝苯地平组和加力+高浓度硝苯地平组,每组再按观察时间点的不同分为5组。用免疫组化方法结合计算机光密度分析系统检测并比较各处理组样本的张力侧与压力侧MMP-13和MMP-10的表达。结果:硝苯地平可以引起正畸牙周膜中基质金属蛋白酶表达的减少,并呈药物浓度增加而减少的更加明显;牙齿受力两侧比较,压力侧变化更显著。结论:硝苯地平阻断Ca2+通道可以使正畸牙周膜中的基质金属蛋白酶表达减少和定位差异减小,即牙周膜成纤维细胞不再对外力产生特异性反应。
Objective To study the effect on expression and localization of matrix metalloproteinase after using the calcium ion channel blocker-Nifedipine during periodontal membrane remodeling. Methods The expression of MMP-13 and MMP-10 on tension side and pressure side were examined by immunehistochemical dyeing and analyzed-with photodensitometry according to different groups. Results The expression of MMP decreased after using Nifedipine,and the decline was more obviously with high concentration of Nifedipine. The impact to pressure side was more significantly. Conclusion Blocking the calcium ion channel by Nifedipine can decrease the MMP expression and diminish the locating difference. Fibroblasts of periodontal membrane can not generate specific reaction to the force.
