摘要
目的建立流式细胞术网织红细胞分析方法,并研究其在临床常见贫血性疾病中的应用价值。方法以噻唑橙作为RNA的荧光染料标记网织红细胞,采用流式细胞仪测定网织红细胞百分数、绝对计数和RNA残留荧光强度。结果健康对照组网织红细胞百分数、绝对计数和RNA残留荧光强度分别为(0.011 4±0.004 0)、(49.75±63.27)、(22.56±34.57)×109/L;溶血性贫血组分别为(4.218 0±0.265 1)、(1 795.25±549.16)、(43.15±48.05)×109/L;缺铁性贫血组分别为(0.027 3±0.0137)、(86.57±67.28)、(23.09±21.00)×109/L;失血性贫血组分别为(3.978 2±1.089 6)、(2 048.01±604.52)、(39.46±88.12)×109/L;慢性肾功能不全组分别为(0.005 2±0.002 5)、(21.59±5.64)、(18.25±9.14)×109/L;肿瘤化疗组分别为(0.020 3±0.022 9)、(24.90±43.35)、(14.27±8.13)×109/L。各贫血组与健康组比较,网织红细胞百分数、网织红细胞绝对计数差异均有统计学意义。结论流式细胞术分析网织红细胞简便快速,重复性好,结果准确可靠,可用于临床各种贫血的诊断与疗效监测。
Objective To establish a method for analyzing reticulacytes by Flow Cytometer and research its clinical applicated values in diagnosing anaemia. Methods Thiazole orange was used as a fluorochrome label to stain reticulocytes by combinding to RNA in reticulcyte plasma. The percent,absolute counting and fluorescence intensity,which images the amount of RNA in reticulcyte plasma,were detected by Flow Cytomter. Results In health control Group,the percent,absolute counting and fluorescence intensity of reticulocyte was (0. 011 4±0. 004 0), (49.75± 63.27) and (22.56±34. 57) ×10^9/L, respectively. In hemolytic anemia group,it was (4. 218 0 ± 0. 265 1), (1 795.25±549. 16) and (43. 15±48. 05)×10^9/L, respectively. In iron deficiency anemia group,it was (0. 027 3±0. 013 7) ,(86.57±67.28) and (23.09±21.00) ×10^9/L,respectively. In blood loss anemia group,it was (3. 978 2± 1. 089 6), (2 048.01 ± 604.52) and (39.46 ± 18.12) ×10^9/L, respectively. In chronic renal insufficiency group, it was (0. 005±0. 002 5) ,(21.59±5.64) and (18. 25±9.14) ×10^9/L,respectively. In tumour chems group,it was (0. 020 3±0. 229), (24. 90±43.35) and (14.27 ± 8.13)×10^9/L, respectively. Compared every anemia group with health control group, the results of percent and absolute counting were obviously different. Conclusion Flow Cytometry analyzing reticulocytes is simple,fast and accurate,and has a good reproducibility. It could use in the diagnosis and curative effect monitoring for various kinds anemia.
