沙利度胺对关节炎伴贫血大鼠血红蛋白的影响

EFFECTS OF THALIDOMIDE ON BLOOD HEMOGLOBIN LEVEL IN EXPERIMENTAL ARTHRITIS RATS ACCOMPANIED WITH ANEMIA

目的探讨沙利度胺对关节炎伴贫血大鼠血红蛋白的影响。方法以胶原诱导性关节炎大鼠作为类风湿性关节炎(rheumatiod arthritis,RA)动物模型,并多次多点皮内注射Ⅱ型胶原及弗氏完全佐剂(CompleteFrund's Adjuvent,CFA)成功诱导出关节炎伴贫血的动物模型。将健康雄性Wistar大鼠76只分为正常对照组、造模组、沙利度胺治疗组,比较3组血红蛋白、大鼠足爪厚度及血清肿瘤坏死因子α(tumor necrosis factor-α,TNF-α)水平及三者之间的关系,探讨RA继发贫血的可能机制及沙利度胺对实验性关节炎伴贫血大鼠的治疗效果。结果大鼠足爪厚度、血清TNF-α水平及血红蛋白浓度间存在密切关系,大鼠足爪越厚关节炎的肿胀程度越高,TNF-α浓度越高;TNF-α浓度越高,贫血也越严重,血清TNF-α浓度与血红蛋白之间呈显著负相关。沙利度胺能够降低关节炎大鼠血清TNF-α浓度,改善关节炎大鼠的贫血情况。结论沙利度胺既能改善关节炎症状又能避免传统抗风湿药物带来的骨髓抑制等不良反应,有望作为新一类的免疫调节剂。

Objective To investigate the effects of thalidomide on blood hemoglobin level in experimental arthritis rats accompanied with anemia. Methods The model of arthritis accompanied with anemia was established successfully through repeat subcutaneous type Ⅱ collagen and Complete Frund＇s Adjuvent（CFA） injection. Seventy-six health male Wistar rats were divided into three groups,the contrast group,the model group and the thalidomide treatment group. Through comparing the relationships of blood hemoglobin concentration, serum tumor necrosis factor-α （TNF-α） concentration and the paw thickness to investigate the possible mechanism of anemia in model rats and the treatment effects of thalidomide on meliorating the anemia symptoms of rats. Results There is a close relationship among hemoglobin concentration, serum TNF-α concentration and the paw thickness,the thicker of the paw and the more severe of the joint swellen,the higher the concentration of the TNF-α and the more severe of the anemia. There is a markedly negative correlation between hemoglobin and TNF-α concentration. Thalidomide can reduce the TNF-α concentration and meliorate the anemia symptoms of collagen induced arthritis rats. Conclusion Thalidomide can meliorate the symptoms of arthritis and avoid the bad effects of anti-rheumatism drugs, it is hopeful to apply thalidomide in the treatment of rheumatiod arthritis patients with anemia.
