摘要
目的探讨核转录因子κB(NF-κB)"诱骗(decoy)"寡核苷酸(ODNs)对结肠癌SW480细胞IL-8表达的影响。方法体外培养SW480细胞,LPS(10μg/L)刺激3h后,用脂质体lipofectin2000介导NF-κB decoy ODNs转染6h,ELISA法检测上清液中IL-8,RT-PCR检测细胞IL-8 mRNA,并设对照组、LPS组、SODN组、lipofec-tin2000组。结果转染NF-κB decoy ODNs细胞的IL-8 mRNA和IL-8的表达高于对照组(P<0.01),但明显低于其他组(P<0.01)。结论NF-κB decoy ODNs明显抑制SW480细胞IL-8的表达。
Objective To study the effect of NF-κB decoy oligodeoxynucleotides on IL-8 expression in LPS-indueed SW480 cells. Methods SW480 cells were cultured in vitro and then stimulated for 3 hours with LPS( 10 μg/L), then added lipofectin2000 mediated NF-κB decoy oligodeoxynucleotides for 6 hours. The supernatants were collected and measured for IL-8 by ELISA; IL-8mRNA was examined by RT-PCR respectively. The results were compared with control group, scrambled ODNs group and lipofectin2000 group. Results After SW480 cells were stimulated by LPS, IL-8mRNA and IL- 8 protein expressions were significantly increased, the difference compared with control group was obvious (P 〈 0.01 ). After NF-κB decoy oligodeoxynucleotides treating , IL-8mRNA and IL-8 protein expressions were significantly inhibited(P 〈 0.01 ) , but the scrambled ODNs group and hpofectin2000 group were not affected. Conclusion NF-κB decoy ODNs can inhibite the expression of IL-8 in SW480 cells.
