低氧对人骨髓间充质干细胞HIF-1α、SDF-1、VEGF mRNA表达的影响 被引量:4

摘要 目的观察低氧对人骨髓间充质干细胞(hBMSCs)缺氧诱导因子-1α(HIF-1α)、基质细胞衍生因子-1(SDF-1)及血管内皮细胞生长因子(VEGF)表达的影响。方法分别在1%O2及20%O2条件下培养hBMSCs,定量RT-PCR技术检测其HIF-1α、SDF-1及VEGF mRNA。结果在1%O2条件下,hBMSCs的HIF-1α、SDF-1、VEGF mRNA表达水平明显增高(P<0.01)。HIF-1αmRNA与SDF-1 mRNA及VEGF mRNA的表达密切相关(r分别为0.488、0.644,P均<0.01),但SDF-1 mRNA与VEGF mRNA的表达无明显关系(r=0.077,P>0.05)。结论低氧可促进hBMSCs的HIF-1α表达,并使SDF-1、VEGF mRNA表达增加。 Objective To observe the effects of low oxygen on the expression of mRNA of hypoxia-inducible factor- 1α(HIF-1α) ,stromal cell derived factor-1 ( SDF-1 ) and vascular endothelial growth factor(VEGF) in hBMSCs. Method The third passages of hBMSCs were cultured in 1% 02 and 20% O2, HIF-1α,SDF-1 and VEGF mRNA were detected by RT-PCR. Results Cultured under 1% O2 condition, the mRNA of HIF-1α,SDF-1 and VEGF of hMSCs increased greatly (P 〈 0.01 ). There was obvious association between the expression of HIF-1 α, SDF-1 and VEGF m RNA( P 〈 0.01 ), but no correlation were found between SDF-1 and VEGF mRNA expression. Conclusion Low oxygen induces the expression of HIF-1α,and results in increasing SDF-1 and VEGF expression in hBMSCs.
