摘要
目的探讨肝细胞癌变过程中cyclinB1的异常表达及其意义。方法用0.05%的2-乙酰氨基芴(2-AAF)诱导Wistar大鼠建立肝癌动物模型,应用免疫组化和原位杂交法检测大鼠肝脏在癌变过程中cyclinB1的蛋白和mRNA表达。结果正常组8只中cyclinB1蛋白和mRNA表达均正常,肝硬化组16只中有12只cyclinB1蛋白和mRNA过表达,肝癌组11例中有4只cyclinB1蛋白和mRNA过表达。结论在3组中cyclinB1蛋白和mRNA表达一致,差异无统计学意义。肝硬化组和肝癌组的cyclinB1异常表达均高于正常组,而且肝硬化组的cyclinB1异常表达高于肝癌组。
Objective To study the expression and significance of cyclin D1 during hepatocellular canceration. Methods 0. 05% 2-AAF induced rat (Wister) hepatocellular carcinoma. CyclinB1 protein and mRNA were examined by immunohistochemistry and in situ hybridization techniques, respectively. Results All rats in control group showed normal expression of cyclinB1 protein and mRNA, 12 of 16 liver cirrhosis cases showed overexpression of CyclinB1 protein and mRNA. Four of eleven hepatocellalar carcinoma cases showed overexpression of CyclinB1 protein and mRNA in the three groups. Conclusion There is no significant difference between expressions of CyclinB1 protein and mRNA. CyclinB1 overexpression in liver cirrhosis and hepatocellular carcinoma shows higher than those in control group. CyclinB1 overexpression in liver cirrhosis shows higher than those in hepatocelluar carcinoma.
