摘要
目的:研究精氨酸-甘氨酸-天冬氨酸(RGD)类短肽对金黄地鼠颊囊癌生长和血管形成的影响,探讨精氨酸-甘氨酸-天冬氨酸-缬氨酸(RGDV)抗肿瘤的作用机制。方法:采用8周叙利亚金黄地鼠,建立二甲基苯丙蒽(DMBA)诱发的颊囊癌模型;运用RGDV进行干预;从整体水平上观察RGDV对肿瘤生长的影响;采用免疫组织化学的方法检测肿瘤中BrdU和vWF-VIII的表达。结果:RGDV具有抑制肿瘤生长的作用,瘤体抑制率达到53.36%;RGDV能抑制肿瘤细胞的增殖、降低肿瘤组织中微血管密度。结论:RGDV可通过抑制肿瘤细胞增殖及肿瘤新生血管形成而发挥抗肿瘤生长的作用。
Objective: To study the inhibition effect of synthetic peptide RGDV on the growth and angiogenesis of hamster buccal-pouch carcinoma and to investigate the antitumor mechanism of RGDV. Methods: Hamster buccalpouch carcinogenesis model was established in 32 hamsters by topical application of 5 g/L DMBA in left buccal pouch three times per week for 12 weeks. The effects of RGDV on the growth of carcinoma were investigated, 5-bromo-2'- deoxyuridine (BrdU) and yon Willebrand factor-VIⅡ (vWF-VIII) expressions were examined by immunohistochemieal staining. Results: RGDV inhibited tumor growth. The tumor growth inhibition rate was 53.36%. The expression of micro-vascular density (MVD) was significantly downregnlated. Conclusion: RGDV shows anti-tumor effects by inhibiting proliferation and angiogenesis of carcinoma.
