一氧化氮合酶基因在慢性肾功能衰竭大鼠阴茎组织表达的价值 被引量:1

摘要 目的探讨慢性肾功能衰竭(CRF)性阴茎勃起功能障碍的发病机制。方法应用SD雄性大鼠同期行5/6肾脏切除术建立CRF动物模型。36只大鼠分为假手术组(NCRF组,n=6)和CRF模型大鼠组(CRF组,n=30),分别于12周注射阿朴吗啡[APO,80μg/(kg.d)]后观察大鼠阴茎勃起情况,应用RT-PCR检测大鼠阴茎海绵体内皮型一氧化氮和酶(eNOS)和神经型一氧化氮和酶(nNOS)基因的表达情况。结果CRF组阴茎勃起率为28%(7/25),NCRF组阴茎勃起率100%(5/5),两组相比,差异有统计学意义(P<0.05)。CRF组阴茎勃起次数为(1.0±0.0),NCRF组阴茎勃起次数为(2.2±0.8),两组相比,差异有统计学意义(P<0.05)。CRF组大鼠阴茎海绵体组织nNOS基因表达低于NCRF组(0.21±0.07 vs 0.40±0.08,P<0.01)。CRF组大鼠阴茎海绵体组织eNOS基因表达低于NCRF组(0.28±0.08 vs 0.67±0.13,P<0.01)。结论慢性肾功能衰竭明显降低大鼠阴茎勃起功能。慢性肾功能衰竭大鼠阴茎勃起功能降低与大鼠阴茎海绵体组织eNOS和nNOS基因表达减少有关。 Objective To investigate the mechanisms of erection dysfunction(ED) induced by chronic renal failure (CRF). Methods CRF experimental models were established by removing 5/6 kidney from male sprague-dawley (SD) rats in one stage. Both the control group (NCRF group, n = 6) and the experimental group (CRF group, n = 30) were, injected with a low dose [ 80 μg/( kg·d) ] of apomorphine (APO) at the 12th week after resection surgeries to measure the correspondent penile erections. Reverse transcriptase polymerase chain reaction (RT-PCR) was used to determine the gene expression of endothelial nitric oxide synthase (eNOS) and neuronal nitric oxide synthase (nNOS) in the rat corpus cavemosum in the 12th week after the resection surgeries. Results The penile erection rado of the CRF group was 28 % (7/25) while that of the NCRF group was100% (5/5) ( P 〈 0.05). The average penile erection frequency of the CRF group was( 1.0 ± 0.0) while that of the NCRF group was (2.2 ± 0.8) (P 〈 0.05). The gene expression of nNOS in the rat corpus eavemosum was (0.21 ± 0.07) of the CRF group and (0.40 ± 0.08) of the NCRF group ( P 〈 0.01 ). The RT-PCR results showed that the gene expression of eNOS in the rat corpus eavemosum of the CRF group was significandy lower than that of the NCRF group[(0.28 ± 0.08) vs(0.67± 0.13) (P 〈 0.01)]. Conclusions CRF signifieandy reduces the erectile function of the rats, and CRF-related ED has some correlations with the decrease of the gene expressions of eNOS and nNOS in the rat corpus cavemosum.
