摘要 热休克蛋白90(HSP90)是一组结构高度保守的蛋白质,作为分子伴侣参与细胞的发育、生长和分化,近年来因HSP90与肿瘤发生发展、生物学行为及其预后有较密切的关系,而成为抗癌药物作用的靶点。很多癌基因蛋白均为HSP90的作用靶点,HSP90抑制剂将促使在肿瘤生长信号通路中起重要作用的HSP90效应蛋白降解,从而阻断肿瘤增殖信号通路的多个靶点,有效阻止肿瘤的生长,体内外实验也证实了HSP90抑制剂的抗肿瘤活性。 Heat Shock Protein 90(HSP90) is a kind of protein with extremely conservation, which participates cell development, growth and cell differerntiation as molecular chaperones. HSP90 is becoming the target of anticancer drugs because of its close relationship with occurrence and development, biological behaviour and prognosis of tumor. Many oncogene proteins are target of effect of HSP90. HSP90 inhibitors will promote the degradation of HSP90 effect proteins, which plays an important role in signaling of tumor growth, and then will inhibit the tumor growth by blocking multiple targets in signaling of tumor proliferation. Experiments in vivo or vitro also testified the anti-tumor activity of Hsp90 inhibitor.
