摘要
目的:研究解偶联蛋白2(UCP2)基因启动子区-866G/A的多态性在中国北方汉族正常人群及糖尿病人群的分布,了解其与2型糖尿病(T2DM)及β细胞功能关系.方法:采用PCR及PCR-RFLP方法检测101名正常人及192名T2DM患者的UCP2基因启动子区-866G/A的多态性,对T2DM患者进行葡萄糖耐量试验,计算β细胞功能指数及胰岛素敏感指数.结果:-866G/A基因型频率在中国北方汉族正常人群及T2DM人群中的分布分别为:GG31例(30.7%),GA46例(45.5%),AA24例(23.8%)及GG44例(28.1%),GA103例(49.6%),AA45例(22.3%),统计分析无显著性差异.②-866G/A多态性的含A等位基因型与胰腺β细胞的胰岛素分泌功能指数ΔI30/ΔG30,HOMAβ,MBCI及INS30呈负相关.与胰岛素敏感指数IAI无关.结论:中国北方汉族人群的UCP2基因启动子区-866G/A基因型分布与日本人相似,与欧洲白人及高加索人群存在显著性差异.②UCP2基因启动子区-866G/A基因型在两组人群的分布无差异.③UCP2基因启动子区-866G/A多态性与胰腺β细胞的胰岛素分泌功能相密切相关.
AIM: To study the distribution of uncoupling protein (UCP) 2 - 866G/A polymorphism in normal subjects and diabetes patients of northern Chinese and to elucidate the association of UCP2 polymorphism with type 2 Diabetes and insulin secretion function of pancreatic β cells. METHODS : The UCP2 - 866G/ A polymorphism was detected by PCR and PCR-RFLP in 101 normal subjects and 192 diabetes patients. 192 diabetes patients underwent 75g glucose OGTTs with measurement of insulin secretion index and insulin sensitive index to study the association of UCP-2 polymorphism with type 2 diabetes and insulin secretion function. RESULTS : (1) The distribution of UCP2 polymorphism in normal subjects and diabetes patients was GG31 (30.7 % ), GA46 (45.5%), AA24(23.8 % ) and GG44(28.1% ), GA103(49.6 % ), AA45 (22.3 % ) respectively. There was not statistical significance. (2) GA and AA of UCP2 - 866G/A Polymorphism were negatively associated with △I30/△G30, HOMA-β, MBCI and INS30. CONCLUSlON:(1) The distribution of UCP 2 -866G/ A polymorphism is similar in Chinese and Japanese, while it is significantly different in Chinese, white Europeans and Cauca- sians. (2) There is no statistical correlation between UCP2 - 866G/A polymorphism and diabetes mellitus. (3) UCP2 - 866G/ A polymorphism is closely associated with insulin secretion function of pancreatic β cells.
