HK-Ⅱ基因的多态性分析及其与原发性肝癌的关系 被引量:1

摘要 目的:探讨已糖激酶-Ⅱ(HK-Ⅱ)基因的多态性位点和原发性肝癌(PHC)易感性之间的关系。方法:应用PCR-SSCP、PCR-RFLP和PCR产物测序技术对80例原发性肝癌患者、122例健康对照者的HK-Ⅱ基因进行序列分析及测序。结果:①HK-Ⅱ基因启动子活性区在病例组和对照组中均没发现多态性位点;②HK-Ⅱ基因第4外显子142位遗传密码存在CAA→CAT多态性改变,在PHC组和健康对照组之间差异有统计学意义(P<0.05);③第15外显子736位遗传密码存在CCC→CCG的多态性改变,产生的SS基因型在病例组和对照组的分布差异有统计学意义(P<0.05);④联合分析发现,SS基因型和第4外显子的多态性改变同时存在的情况在两组之间差异有统计学意义(P<0.05)。结论:①HK-Ⅱ基因的启动子活性区碱基组成稳定,不易发生突变;②CAA→CAT和CCC→CCG的多态性改变会增加PHC的发病危险;③SS基因型和CAA→CAT多态性改变同时存在时,会增加个体患肝癌的危险性。
