巨噬细胞刺激人胃癌MGC803细胞生长与促血管生成活性的研究

摘要 目的:研究人白血病单核巨噬细胞THP-1对人胃低分化黏液腺癌MGC803细胞生长与促血管生成因子VEGF表达的影响。方法:通过流式细胞仪(FCM)测定细胞周期的改变,RT-PCR检测MGC803细胞VEGF表达情况。结果:免疫细胞化学分析发现,与对照组比较,处理组VEGF表达明显增强,其中以非直接联合培养组为最强;RT-PCR检测发现,MGC803细胞能表达VEGF mRNA4种剪切体VEGF121、VEGF145、VEGF165、VEGF189;各处理组4种剪切体表达均有增强,且都以非直接联合培养为最强。流式细胞仪检测处理组MGC803细胞S期细胞百分比均有增加,其中以非直接联合培养组最高。结论:THP-1细胞条件培养基处理、与THP-1细胞非直接联合培养能促进MGC803细胞的增殖活性及细胞周期G1-S期转变。MGC803细胞能表达VEGF mRNA的4种剪切体;THP-1细胞条件培养基处理、与THP-1细胞非直接联合培养能促进MGC803细胞VEGF表达。
