摘要
本实验选用Wistar新生大鼠,出生后第2天开始腹腔注射葡萄糖或葡萄糖加精氨酸共6天,每日2次,对照组不做处理.分别于生后第10、20天剖腹取胰腺,制作光电镜标本,并用免疫组织化学PAP标记β细胞.同时测定β细胞及其分泌颗粒的数面密度(NA)和体密度(Vv).结果显示:对照组第20天胰β细胞及其分泌颗粒的NA、Vv均明显高于第10天组;尽管实验组β细胞的NA、Vv低于对照组.但其β颗粒的NA、Vv却显著高于对照组。上述结果提示.大鼠生后发育期间胰岛β细胞及其分泌颗粒的数量、体积逐渐增加,而葡萄糖、精氨酸主要引起β细胞提前成熟。
Newborn Wistar rats were treated twice a day for 6 days after birth by glucose (G) or glucose with arginine (G + A ).The animals were killed on the tenth and twentieth day of life.The pancreas was observed by light and electronic mi-croscopy. The island β celIs were showed by the immunochis-tochemistry (PAP method) and its NA and Vv were measured.The NA and Vv of 6 granules were measured too. The result showed that the NA. Vv of-β cell and β granules were higer in normal rat on the twentieth day than the tenth day. A re markable rise in NA Vv of 6 granules were observed in G and G+A treated rat. It seemed that G and G+A could accelerate β granules maturation.
