摘要
目的:研究氨基糖苷类抗生素所致的豚鼠听力损伤与其耳蜗毛细胞凋亡之间的关系。方法:将36只豚鼠随机分为生理盐水对照组、庆大霉素组和奈替米星组,连续给药14d,利用TUNEL法原位检测动物耳蜗毛细胞的凋亡情况。结果:生理盐水对照组豚鼠耳蜗未见凋亡毛细胞,庆大霉素组豚鼠耳蜗基底回内外毛细胞均消失,第3回中第1排外毛细胞消失,第2,3排外毛细胞发生凋亡,第2回中第1,2排外毛细胞发生凋亡。而奈替米星组豚鼠耳蜗基底回中第1,2排外毛细胞丢失,第3排外毛细胞发生凋亡,第3回中第1排外毛细胞丢失,第2排外毛细胞发生凋亡。第2回毛细胞中仅见第1排外毛细胞凋亡。结论:给予豚鼠庆大霉素或奈替米星后均可引起其听力损伤;其耳毒性的产生可能与其引起的耳蜗毛细胞凋亡密切相关。
OBJECTIVE To study the relation between heating impairment of guinea pig resulted from Aminoglycoside antibiotics and cochlear hair cells apoptosis. METHODS Thirty-six guinea pigs were randomized to normal saline group , Gentamicin group and Netilmicin group. After 14 days' administration consecutively, detect apoptosis of their cochlear hair cells by TUNEL method in situ. RESULTS In normal saline group apoptosis of hair cells were absent, but in Gentamicin group the disappearance Of OHCs and IHCs was observed in the cochlear basal turn and OHCs in the first row of the third turn were disappeared, labeled nuclei could be seen in OHCs in both the second row of third turn and the third row of the third turn, as well in OHCs in the first and second row of the second turn. In Netilmicin group the disappearance of OHCs was observed both in the first and second row of the cochlear basal turn and in the first row of the third turn, labeled nuclei could be seen in OHCs in both the third row of basal turn and the second row of the third turn, as well in OHCs in the first row of the second turn. CONCLUSION Gentamicin and Netilmicin can induce the hearing impairing of guinea pig,which maybe closely related to apoptosis of guinea pig cochlear HCs.
