南方鲇肾细胞系(SMK-Ⅰ)的建立及其细胞骨架的研究 被引量:3

Establishment of a Kidney Cell Line (SMK-Ⅰ) of Southern Catfish (Silurus meridionalis Chen) and Research of Its Cytoskeleton

摘要 南方鲇肾细胞系(SMK-Ⅰ)在培养初期为成纤维样和上皮样细胞的混合细胞系,随着传代次数的增加,形成了稳定的成纤维型细胞系;SMK-Ⅰ细胞的生长周期为7~8天;大多数细胞的染色体为二倍性,也观察到染色体多倍化和异倍化的细胞群体;流式细胞仪分析不同代龄的细胞周期,发现该细胞系G2-M期细胞的数量,随着代龄的增加,逐渐增多,说明该细胞系已经成功转化;Pena法显示其细胞骨架网络比较分散,且随细胞贴壁的情况和本身的形态而异. AbsA continuous cell line was established from the kidney of Southern catfish (Silurus meridionalis Chen) fry. The primary cells were originally a mixture of both epithelial cells and fibroblasts and, with the increase in generations, they became stabilized fibroblastic cell lines and were named "S. meridionalis kidney (SMK-Ⅰ) cell line". The cell growth cycle lasted 7--8 days. SMK-Ⅰ cells were largely diploids, though a small percentage of polyploids and alloploids were also observed. The number of the cells in G2- M stage increased with the generation age, suggesting that SMK-Ⅰ had been successfully transformed. PENA analysis showed that the cytoskeleton framework in SMK-Ⅰcells was considerably scattered and varied with the conditions of the adherent cells and the cell features.
