摘要
用不同质量浓度的秋水仙素液体、固体和固液双层介质处理预培养时间不同的兰考泡桐二倍体幼苗叶片,叶柄和茎段后,在不同培养基上进行体外植株再生,得到了兰考泡桐的四倍体植株.结果表明,固液双层介质处理兰考泡桐叶片四倍体诱导率最高可达23.4%.显微镜观察诱变植株根尖染色体数为2n=4x=80,四倍体幼苗较二倍体幼苗叶片大而厚、叶色深,气孔大,生长更为健壮.
Tetraploid seedlings of Paulownia elongata were obtained from its diploid pre-cultured leaves,petioles and internodes of the plant which were treated on liquid,solid and liquid-solid media with different concentrations of colchicines for various periods.The results indicated that the explants with the highest frequency(23.4%) of tetraploidy on liquid-solid media were its diploid leaves.Chromosome number of the root tips is 2n=4x=80,leaves of tetraploid seedlings are larger,thicker and greener,and the stomata are larger than those of diploid ones.
