摘要
目的研究正常 Sprague-Dawley(SD)大鼠咽鼓管上皮水通道蛋白(aquaporins,AQPs)的分布特点。方法以 SD 大鼠为实验动物,逆转录聚合酶链反应(reverse transcription polymerase chain reaction,RT-PCR)从基因水平检测咽鼓管上皮 AQPs mRNA 的表达,免疫组织化学方法从蛋白水平检测 AQPs 在咽鼓管上皮的分布特点。结果基因水平检测到 SD 大鼠咽鼓管上皮存在 AQP1,3,4,5,免疫组织化学二氨基联苯氨染色法显示 AQP1分布在咽鼓管上皮的毛细血管内皮细胞和成纤维细胞,AQP3,4,5则分别分布于咽鼓管基细胞或无纤毛细胞,纤毛柱状上皮和腺体细胞。结论咽鼓管上皮上分布有 AQP1,3,4,5,各自协同作用共同维持咽鼓管的正常生理功能。(中国眼耳鼻喉科杂志,2007,7:353-355)
Purpose To investigate the distribution of aquaporim (AQPs) in eustachian epithelium in normal rat. Methods Reverse transcription polymerase chain reaction (RT-PCR) analysis and immunohistochemical technique were used for expression of mRNA and protein of AQPs respectively in rat eustachian tube. Results The expression of AQP1,3,4,5 in rat eustachian tube epithelium was confirmed by RT-PCR and immunohistochemical examination. The immunohistochemical analysis (diaminobenzidin method) demonstrated that AQPI was prominent in the capillary endothelium cells and fiberoblasts of the eustachian tube epithelium. AQP3,4,5 were found in the basal cells/non-ciliated cells, ciliated cells and glandular epithelium respectively. Conclusions AQP1,3,4,5 are expressed in the eustachian tube epithelium, which play a vital role in maintaining the normal physiological function of the eustachian tube.
