摘要
目的观察异甘草素(ISL)和Flutamide联合应用对人前列腺癌细胞体外增殖的抑制作用。方法采用MTT法测定细胞增殖,流式细胞仪检测细胞凋亡,逆转录聚合酶链反应(RT-PCR)分析Bcl-2表达。结果ISL和不同浓度Flutamide联合用药比单独应用Flutamide对前列腺癌PC-3细胞增殖抑制作用明显增强(P<0.01),同时显著下调Bcl-2基因表达,增加癌细胞凋亡。结论ISL和Flutamide联合应用对前列腺癌PC-3细胞增殖抑制作用明显增强,其机制可能与下调Bcl-2基因表达及增加癌细胞凋亡有关。
Objective To study the effect of isoliquiritigenin(ISL) liposome combined with Flutamide on the proliferation of human prostate cancer cells in vitro, Methods The cell proliferation was evaluated by MTT method, Cell cycle and apoptosis were determined by flow cytometer(FCM) and RT-PCR was used to detect the expression of Bc1-2. Results ISL combined with Flutamide showed a significantly stronger inhibitory effect on PC-3 cell in vitro than Flutamide alone(P〈0.01), The expression of Bc1-2 was down-regulated and the apoptosis of PC-3 cell was increased also. Conclusion Proliferation inhibitory effect against PC-3 cell in vitro can be enhanced by ISL combined with Flutamide. The possible mechanism is associated with increasing apoptosis of PC-3 cell and decreasing expression of Bc1-2.
