西索米星抗生素的化学发光衍生技术研究

Investigation on the Labelled Reaction for the Chemiluminescence Determination of Sisomicin Antibiotic

研究了N-4-氨基丁基-N-乙基异鲁米诺-N,N'-二琥珀酰亚胺碳酸盐(ABEI-DSC)与西索米星(Sisomicin)抗生素的衍生技术,建立了西索米星抗生素的化学发光衍生技术。研究了ABEI-DSC和ABEI-DSC-Sisomicin体系的化学反应动力学,确定了反应时间、配比、温度等因素对化学发光的影响,建立了西索米星的化学发光标记技术,并应用于Sisomicin抗生素的测定,检出限(S/N=3)达到5×10-7mol/L,为西索米星抗生素残留分析提供了新的研究方法。

A labelled reaction for the chemiluminescence determination of sisomicin antibiotic has been developed by using N-（4-aminobutyl）-N-ethylisoluminol（ABEI）and N,N-disuccinimidyl carbonate（DSC）.Characteristics of the systems of ABEI-DSC and ABEI-DSC-sisomicin were investigated.In this work,the chemiluminescence dynamics of ABEI-DSC-sisomicin was achieved,and effects of the factors such as reaction time,the proportion of ABEI-DSC or ABEI-DSC-sisomicin and reaction temperature were surveyed.Under the optional conditions,the labelled reaction were formed and up to the maximum after 4 hours under ultrasound.In the analysis of sisomicin,a satisfactory result was obtained with the limit of detection（S/N=3）of 5×10^-7mol/L.
