摘要
以明胶为原料,用悬浮成球、甲苯制孔的方法制备大孔明胶微载体。考察了表面活性剂及其配比、油水比、明胶浓度、制备方式和表面修饰等对载体性能的影响,优化了制备大孔明胶微载体的条件,制得了直径为100~300μm,孔径为10~30μm的大孔明胶微载体。
Macroporous microcarriers are new support materials with important advan tages in both industrial cell culture processes and the culture of cells of medical significance. Using gelatin as raw materials and toluene as oil phase medium, a new kind of macroporous microcarriers was developed by the method of oil in water in oil emulsion. Their diameter and pore ranges are 100 ̄300μm and 10 ̄30μm, respectively. A novel strategy was adopted to overcome the poor interconnection of pores in macroporous microcarriers, which is a key parameter of porous carriers. The effects of different surfactants (Tweens and/or Spans), ratio of oil to water and preparation procedures on the features of gelatin based macroporous microcarriers were investigated and the optimized conditions were selected.
