摘要
目的:研究补充赖氨酸对力竭运动大鼠心、肝细胞凋亡的影响,观察其对心、肝细胞凋亡调控基因Bax和Bcl-2的变化,探讨赖氨酸对力竭运动大鼠机体的保护作用。方法:成年健康雄性SD大鼠24只随机分成正常对照组、运动组和给药组。正常对照组不运动,灌胃生理盐水1次;运动组进行灌胃生理盐水1次,而给药组进行灌胃L-赖氨酸1次,运动组和给药组的运动强度依Bedford所建立的运动负荷模型。实验结束用免疫组织化学法检测心、肝细胞凋亡调控基因Bax和Bcl-2的阳性物质灰度值、面积和含量。结果:与对照组相比较,运动组和给药组心、肝细胞凋亡调控基因Bax灰度值差异显著,具有显著性意义(P<0.05),与运动组相比较,给药组也具有显著性差异(P<0.05);与对照组相比较,运动组的心、肝细胞凋亡调控基因Bcl-2灰度值具有显著性意义(P<0.05),与运动组相比较,给药组心肌细胞凋亡调控基因Bcl-2灰度值也具有显著性意义(P<0.05)。结论:大强度急性力竭运动可使大鼠心、肝组织致损伤,诱导心、肝细胞凋亡,L-赖氨酸对急性力竭运动大鼠心、肝组织有保护作用。
The purpose of this study was to analyze the effect of lysine supplement on exhaustive rat's liver and heart apoptosis bax and bcl-2 gene and discuss the protection of lysine on mice after exhausting exercises. 24 SD rats were divided into 3 groups randomly: control group (C) , exhausting exercise group (EE) and exhausting exercise with supplying lysine (EESL). Control group has no exercise with feeding water one time ,exhausting exercise group was fed with water one time and the exhausting exercise with supplying lysine was fed with L-lysine one time. The sports burden model of Bedford was established in order to test the liver and heart apoptosis Bax, Bcl-2 gene with immunohistochemisty technology and gain the GREYLEVEL, AREA and CONTENT. The result showed that exhausting exercises can induce apoptosis, supplying lysine to exhausting exercise's rats can decrease liver and heart apoptosis Bax gene(P〈0.05). Compared EESL with EE, there has some changes about Bax gene vitality in rats(P〈0.05). It decreased liver and heart apoptosis Bcl-2 gene(P〈0.05). Compared EESL with EE, there has some changes about Bcl-2 gene vitality in rats(P〈0.05). It concluded that exhausting exercises can lead damage on liver and heart and induce liver and heart apoptosis. It approved that supplying lysine can protect liver in exhausting exercises.
