人前列腺癌PC-3M细胞中与转移相关靶标蛋白的筛选 被引量:2

摘要 目的探讨早期诊断前列腺癌和逆转前列腺癌转移的标志物。方法采用一维SDS—PAGE结合高效液相色谱-串联质谱的方法分离并鉴定PC-3M细胞中的膜蛋白。结果在严格的过滤参数条件下(当Charge+1,Xcorr≥1.9;当Charge+2,Xcorr≥2.2;当Charge+3,Xcorr≥3.75;其中DelCN≥0.1),PC-3M细胞中鉴定出2023个蛋白,其中1391(69%)个蛋白经过基因本体评注(GOA)显示为已知细胞组分,其余为未知细胞组分。在已知细胞组分中527(38.30%)个蛋白为膜蛋白或者膜相关蛋白,其中18.32%的蛋白被预测至少有1个跨膜区,49.62%的蛋白有1~3个跨膜区,19.08%的蛋白有4~9个跨膜区,9.92%蛋白预测有≥10个跨膜区。除了已知的与膜相关的蛋白,很多新的蛋白也被鉴定,其中包括假设蛋白和一些cDNA序列。结论一种明显独特的质膜蛋白表达模式存在于高转移前列腺癌细胞系中。这将帮助进一步理解前列腺癌侵袭和转移的分子机制,并且为寻找前列腺癌转移相关的靶标分子提供实验基础。
