摘要
目的探讨外周血单个核细胞(mononuclearcells)NADPH氧化酶(NADPH oxidase)活性及表达与颈动脉硬化的关系。方法采用高频超声测定入选者颈动脉内-中膜厚度(carotidintima-media thickness,IMT),分为IMT正常组(n=28)及IMT增高组(n=32),记录年龄、血压、血糖及血脂各参数,以血清丙二醛(malondialdehyde,MDA)和蛋白羰基化水平比较两组氧化应激(oxidative stress)状态,并用RT-PCR检测NADPH氧化酶核心酶NOX2表达,间接免疫荧光方法检测其活性。结果颈动脉硬化组血清丙二醛增高(P<0.05),蛋白羰基化水平高(P<0.05),NOX2亚单位表达高(P<0.05)。结论单个核细胞NADPH氧化酶活性与动脉硬化发生发展有密切关系,氧化应激促进动脉硬化(atherosclerosis,AS)。
Objective To investigate the correlation between NADPH oxidase in peripheral mononuclear cells (PMNCs) and carotid intima-media thickness (IMT) , and the impact of oxidative stress on atherosclerosis (AS). Methods Carotid IMT of the subjects was determined by high-frequency ultrasound. The subjects were divided into normal IMT group ( n =28 ) and increased IMT group ( n = 32) to record age, blood pressure, glucose and lipid. The oxidative stress status was compared between two groups by serum malondialdehyde (MDA) and protein earbonylation levels. The expressions and activity of NOX2, core enzyme of NADPH oxidase were detected by RT-PCR and indirect immunofluorescence respectively. Results The levels of serum MDA and protein oxidative carbonyl, NOX2 subunits expressions in increased IMT group were significantly higher than those in normal group ( P 〈 0. 05 ). P47phox transferred into cellular membrane. Conclusions NADPH oxidase activity of PMNC has close relation to AS. Oxidative stress could promote AS.
