摘要
目的研究1-磷酸鞘氨醇(S1P)对豚鼠心室肌细胞动作电位(AP)及电压依赖性钾电流(KV)的作用及其机制。方法利用Langendorff离体心脏逆向灌流法分离豚鼠心室肌细胞,采用全细胞膜片钳技术记录心室肌细胞AP和KV。结果S1P可延长动作电位复极50%时程(APD50)和动作电位复极90%时程(APD90),降低KV,其作用可被百日咳毒素(PTX)和卡弗他丁C(Calphostin C)阻断。结论S1P可能通过PKC途径抑制KV及延长AP时程。
Objective To investigate the effect and mechanism of sphingosine 1-phosphate (S1 P) on action potential (AP) and voltage-dependent potassium current (Kv)in isolated guinea pig ventrieular myocytes. Methods The yentricular myocytes were isolated by using Langend patch-clamp recording technique. Results S1P orff perfusion method. The AP and Kv were recorded by whole cell prolonged the 50% and 90% action potential duration (APDso and APDgo) and decreased Kv which were blocked by pertussis toxin (FIX) or Calphostin C. Conclusion SIP decreased Kv, in a PKC pathway and prolonged action potential duration in guinea-pig ventrieular myoeytes.
