急性早幼粒细胞白血病TNF-α基因多态性和蛋白表达水平的相关研究

摘要 目的探讨北方地区汉族人肿瘤坏死因子-α(TNF-α)基因型别的多态性和TNF-α蛋白表达与急性早幼粒细胞白血病(APL)的相关性。方法对21例APL患者采用酶联免疫分析法(ELISA)测定TNF-α含量,应用顺序特异性引物聚合酶链反应(PCR-SSP)方法检测TNF-α基因型别的多态性变化,并与31名健康献血者对照。结果APL组血中TNF-α蛋白含量(1.810±0.116)显著高于对照组(1.107±0.095),P<0.05;APL组和正常对照组TNF-α(308A)等位基因频率分别是(9.524%)和(9.68%)组间差异无统计学意义(P>0.05),提示该等位基因频率与APL不相关,血中TNF-α蛋白表达增高与APL相关。结论中国北方地区汉族人TNF-α基因308位点基因多态性与APL的易感性、临床以及预后可能不起重要作用;而TNF-α蛋白表达与APL的易感性相关联。 Objective:To observe the expression levels and gene polymor-phisms of TNF-α in peripheral blood mononuclear cells of patients with acute promyelocyti leukemia(APL);to study the effects of the expression levels and gene polymorphisms of TNF-α in the patients with APL.Methods: We select peripheral blood of 21 patients with APL and 31 controls.We use enzyme-linked immunosorbent assay(ELISA) to measure the expression levels of TNF-α in serum;and by using polymerase chain reaction and sequence specific primers(PCR-SSP),gene polymor-phisms of TNF-α in mononuclear cells are analyzed.Results: The TNF-α levels were higher in APL patients than controls(P〈0.05);The frequency of TNF-α(308A) genotype was not significantly higher in APL patients than that of controls(P〉0.05).Conclusion: Our results indicate that the expression levels may be related to development of APL.No significant difference was found in allele frequency at TNF-α(308A) positions.
