黄芪甲苷对大鼠心肌基因表达谱的影响 被引量:10

摘要 目的:观察黄芪甲苷连续给药对大鼠心肌基因表达谱的影响。方法:雄性Wistar大鼠以黄芪甲苷5mg.kg-1.d-1腹腔注射,连续2周。对照组给予相同容积的黄芪甲苷溶剂。取黄芪甲苷给药组大鼠心肌组织和相同部位的溶剂对照组大鼠心肌组织进行基因芯片对照检测,扫描仪扫描芯片荧光信号,用相应软件分析差异表达基因和基因功能。结果:黄芪甲苷组显示72个上调基因,其中39个基因生物学特性明确。经基因功能分析,黄芪甲苷对血管发育功能的影响最大;其次是对心肌发育功能和与细胞骨架表达及细胞收缩相关功能有影响。结论:黄芪甲苷对促进血管和心肌发育、增强心肌功能等基因上调的作用最为明显。 Objective: To observe effect of astragaloside Ⅳ on gene expression profile of rat myocardium. Method: Astragalosid Ⅳ 5 mg · kg^-1· d^-1 was administrated intraperitoneally to male Wistar rats for 2 weeks and control rats were given same volume of solvent for astragaloside Ⅳ. Left ventricular myocardium following astragaloside Ⅳ treatment and the same regions of control group were used for gene microarray analysis. The cDNA microarray was scanned for the fluorescent signals. Differential expression genes and gene function were analyzed using related software respectively. Result: Seventy two genes were up-regulated by astragaloside Ⅳ significantly. Among them, biological characteristics of 39 genes were definite. After analysis on their function, the results showed that the effect of astragaloside Ⅳ on blood vessel development genes was most markedly up-regulated and its enhancement on genes related with cardiac muscle development, cell skeletal protein expression and cell contraction were also significant. Conclusion: Astragaloside Ⅳ could improve genes related to development of blood vessel and myocardial contraction in rat left ventricle.
