摘要
研究雷公藤治疗系统性红斑狼疮的作用机制。以激活的正常人外周血单个核细胞(PBMC)为细胞模型,应用荧光激活细胞分类仪(FACS)分析雷公藤单体TZ93对PBMC细胞表型的影响。10名正常女性的PBMC分成3组:Ⅰ组PBMC,Ⅱ组PBMC+PHA,Ⅲ组PBMC+PHA+TZ93,分别加入鼠抗人CD8、CD4、CD25、NK抗体。结果:FACS显示TZ93能显著抑制激活的PBMC中CD4+、CD8+、CD25+的表达及NK活性。结论:TZ93能明显抑制PBMC表型的表达,是一种有效的细胞免疫抑制剂。
In order to further investigate the mechanism of action of Tripterygium wilfordii Hook(TWH, a kind of Chinese herbs) on systemic lupus erythermatostis (SLE) and other autoirmmune diseases.METHODS We cultured phytohemagglutinin (PHA)-activated normal peripheral blood mononuclear cells (PBMC) in the presence of TWH compound TZ 93. Each sample was divided into three groups: Ⅰ: PBMC only, Ⅱ: PBMC+PHA,Ⅲ: PBMC+PHA+TZ 93. Tritiated thymidine incorporation assay, fluoresenee activating cells sorter were used.RESULTS TZ 93 at concentration of 10 mg/ L, could inhibit the proliferation of PBMC, with no evidence of direct cytotokicity. The value of CD4, CD25, NK in group Ⅲ decreased compared with those in group Ⅱ respectively.Interestingly, there was no difference in CD25 between group Ⅰand Ⅲ.CONCLUSIONS TZ 93 can inhibit expression of CD4 CD25, NK Cell in PHA--activated PBMC, especially has better effeCt on expresssionod cd25 So, our findings suggested that CD4, CD25 and NK cell mediated mechanism for suppression of lymphocyte prolderation by TZ 93, which might account for the inununosuppressive effects TWH in Patients with SLE.
