摘要
目的:建立测定阿昔洛韦的光度法。方法:阿昔洛韦与固绿FCF(FastgreenFCF)染料反应,形成离子缔合物,溶液颜色发生明显改变,最大褪色波长在632nm,同时在598nm处产生正吸收。在最大褪色波长处,阿昔洛韦的浓度与溶液的褪色程度呈良好线性关系,可用分光光度法测定。结果:在最大褪色波长632nm处,阿昔洛韦的浓度在4.65×10-7~1.02×10-5mol.L-1范围内遵守比尔定律,表观摩尔吸光系数ε632为2.25×104L.mol-1.cm-1,检出限为2.93×10-7mol.L-1。结论:方法具有较高的灵敏度和良好的选择性,用于实际药品、血浆及尿液中阿昔洛韦的测定,结果满意。
OBJECTIVE A sensitive spectrophotometric method is developed for determination of aciclovir. METHODS Aciclovir reacts with fast green FCF to form an ion association complex, the colors of solution changes obviously, with its nega tive absorption maxima at 632 nm and positive absorption maxima at 598 nm. The fading colour imensity is also proportional to the concentration of aciclovir, spectrophotometric method is applied to the determination of aciclovir. RESULTS The concentration of aciclovir in the range of 4. 65 ×10^-7 ~1.02 ×10 s mol.L^-1 obeys Beefs law at the negative absorption maxima at 632 nm, the apparent molar absorptivity ε632 is 2. 25× 10^4 L·mol^-1·cm^-1 , the detection limit is 2. 93 ×10^-7 mol. L^-1. CONCLUSION The method has high sensitivity and fairly good selectivity, and can be applied to the determination of aciclovir in medi- cine samples and human serum and human urine, with satisfactory results.
