决明子袋泡剂及其特征图谱考察

Study on the specific chromatogram of the Cassiae obtusifolia Teabag

目的:探求决明子袋泡剂的高效液相色谱特征,为决明子袋泡剂的生产、使用提供特征图谱依据。方法:样品加入沸水35mL,置盖保温,浸出液加盐酸4.7mL,加热水解,用氯仿回流提取。采用梯度洗脱技术进行高效液相分析。色谱柱为ZorbaxEclipseXDB-C18柱,流动相为乙腈-0.1%磷酸水溶液,体积流量为1mL.min-1;检测波长为278nm,参比波长为550nm。用数字化色谱综合信息指数法处理色谱特征信息。结果:得到了不同浸出时间的决明子袋泡剂提取液的特征图谱,且浸出时间不同,决明子袋泡剂提取液的检出峰数、检出率、重叠峰数、重叠率有较大差别,经综合信息指数法比较相似度,决明子袋泡剂保温20min提取液(11～20号)的相似度低于决明子袋泡剂保温40min提取液(1～10号)的特征图谱库的相似度阈值。结论:采用特征图谱可控制决明子袋泡剂在不同保温时间下的内在质量。

OBJECTIVE To investigate HPLC profiles of the Cassiae obtusifolia Teabag and provide basis for the production and use of specific chromatogram of Cassiae obtusifolia. METHODS Boited water was added to the samples of Cassiae obtusifolia, covered and heated. The lixivium was hydrolyzed with hydrochloride acid and then reflux extracted with chloroform. The HPLC was conducted on Zorbax Eclipse XDB-C18 column with acetonitrile-water （0. 1% phosphoric acid） as mobile phase in gradient elution at a flow rate of 1.0 mL. min^- 1. The detection wavelength and reference wavelength were 278 nm and 550 nm, respectively, The integrated chromatographic-digitized information was treated by index of composite information. RESULTS The specific chromatograms of the Teabag lixivium in different times were obtained and the detecting peaks and ratio, overlapping peaks and ratio of the Cassiae obtusifolia Teabag are different at different times, To compare the similarity by index of composite information, the lixivium heated for 20 min was less than that for 40 min. CONCLUSION The quatity of Cassiae obtusifolia Teabag could be controlled by this methed.
