虎杖甙对内毒素刺激的单个巨噬细胞内游离钙动态变化的影响 被引量:8

摘要 用荧光染料Indo-1标记小鼠腹腔巨噬细胞内游离钙,在粘附式细胞仪检测下观察细胞内游离钙的动态变化,在大肠杆菌O55:B5内毒素刺激下细胞内游离钙迅速升高,于第400s达峰值.经虎杖甙处理的巨噬细胞加入内毒素后,胞质游离钙无明显增高.因细胞浴于无钙环境,故胞内游离钙升高不是由于细胞外的Ca2+进入细胞,而是由于内毒素影响胞内钙库调节功能.虎杖甙可能保护胞内钙库中胞质钙的某种调节机制,从而避免胞质游离钙升高. The free calcium in single mouse peritoneal macrophages was labelled with fluorescence dye Indo-1 and its dynamic changes were observed with Adherent Cell Analysis and Sorting Cytometer (ACAS)570. The level of intracellular free calcium elevated rapidly after the stimulation of lipopolysaccharide (LPS) from E. coli O55: B5 with a peak at 500 s. After the addition of LPS into the macrophages processed with polydatin,no obvious elevation of intracellular calcium ion level could be seen. Because the cells were in a calcium-free environment,intracellular Ca2+ elevation was unlikely due to extracellular calcium influx,while LPS interfered with the regulation of calcium pool within the macrophages. Polydatin might protect some regulatory mechanisms of cytosolic Calcium in intracellular calcium pool,and so avoid cytosolic Ca2+ elevation.
