颊癌中肿瘤转移抑制基因nm23mRNA的表达 Ⅰ.Northern斑点杂交研究 被引量:4

摘要 通过Northern斑点杂交技术研究52例颊癌及相关组织中肿瘤转移抑制基因nm23-H1和nm23-H2mRNA的表达。结果发现:颊癌组织中nm23-H1和nm23-H2mRNA表达比正常颊粘膜、白斑、癌旁粘膜均有不同程度增高。颊癌有转移组中nm23-H1mRNA表达比无转移组明显降低,转移灶中nm23-H1mRNA的表达更低(P<0.05)。11例有转移颊癌患者中有9例(81.8%)为nm23-H1低表达;而19例无转移颊癌,有15例(78.9%)为高表达(P<0.05)。nm23-H2mRNA表达在有无转移组患者中无明显差异(P>0.05)。结果提示,在颊癌的转移过程中,nm23-H1比nm23-H2起着更重要的作用。nm23-H1mRNA表达可作为预测有无颊癌淋巴结转移的一个有价值的指标。 In this study the authors detected both nm23 H 1 and nm23 H 2 mRNA levels in 52 tissues samples of patients with carcinoma of buccal mucosa (CBM) by Northern blotting method. The expression levels of both nm23 H 1 and nm23 H 2 mRNA in CBM were higher than those in normal buccal mucosa, leukoplakia, adjacent nontumorous mucosa, and lymphonodes with or without metastasis. The nm23 H 1 mRNA expression levels in CBM with lymphonodes metastases were lower than those in CBM without metastasis, and were lower in metastatic lymphonodes (P<0.05). Of the 11 cases with metastases, 9 cases (81.8%) showed low expression of nm23 H 1 mRNA, of 19 cases without metastasis, 15 cases (78.9%) were high expression. No significance of nm23 H 2 mRNA expression levels were found between CBM with and CBM without metastasis (P>0.05). The expression of nm23 H 1 and nm23 H 2 mRNA showed no significant correlation with each clinical pathological parameter examed. The result suggested that the expression of nm23 H 1 mRNA significantly correlated inversely with lymphonode metastasis in CBM, while the expression of nm23 H 2 mRNA not. nm23 mRNA expression levels can provide a valuable predictor of lymphonode metastases in CBM.
