摘要
目的建立反相高效液相色谱法测定宁夏枸杞中绿原酸含量的方法。方法色谱分析条件Shim-pack VP-ODS色谱柱(250mm×4.6mm,5μm),以乙腈∶水∶冰乙酸(5∶94.5∶0.5)为流动相A,乙腈∶水∶冰乙酸(70∶29.5∶0.5)为流动相B,采用梯度洗脱,流速0.5ml/min,检测波长342nm,柱温30℃。结果绿原酸线性范围为10~100μg/ml(相关系数为r=0.9995),平均回收率为98.6%(n=5);不同产地、不同年份宁夏枸杞样品中绿原酸含量存在差异,随保存时间的延长,绿原酸含量减少,但减少程度不明显。结论本方法重现性好,适用于测定枸杞中绿原酸的含量。
Objective To develop a RP-HPLC method with gradient dilution for determination of chlorogenic acid in Lycium barbarum.L from different habitats.Methods The separation was performed on a Shim-pack VP-ODS 250 mm ×4.6 mm(5 μm),acetonitrile/water/acetic acid(5 ∶ 94.5 ∶ 0.5)was used as the mobile phase A and acetonitrile/water/acetic acid(70 ∶ 29.5 ∶ 0.5)as the mobile phase B with gradient elution,the flow rate was 0.5 ml/min,the detective wavelength was at 342 nm and the temperature was 30℃.Results The good linearity of chlorogenic acid was obtained over the ranges of 10~100 μg/ml(r=0.999 8),the average recovery was 98.6%(n=5); the content of chlorogenic acid in Lycium barbarum.L from different habitats was different.Conclusion Being rapid and reproducible,the method can be used to determine chlorogenic acid in Lycium barbarum.L.from different habitats.
