摘要
目的:评价人IL4cDNA转染胃癌细胞株分泌IL4的可行性和γ射线照射的安全性。方法:用脂质体转染法将人IL4cDNA转入SGC-7910人胃癌细胞株,并检测细胞分泌的IL4和50Gyγ射线照射对肿瘤细胞分泌的影响。结果:人IL4cDNA可以转染入人胃癌细胞,并表达有活性IL4。基因转染可以明显增强IL2激活的LAK细胞的肿瘤杀伤作用。用大剂量的γ射线照射后,转染细胞仍可以持续分泌IL43-4周。结论:人IL4cDNA转染不改变胃癌细胞的体外生长特性,但可以明显增强抗肿瘤免疫反应。大剂量γ射线照射可能是IL4基因治疗的安全措施。
Objective: To evaluate the possibility of continuous production of IL 4 by a human IL 4 gene transfected human gastric cancer cell line (GCC) and the safety of γ ray irradiation. Methods: After the human IL 4cDNA was transfected into the SGC 7901 line with the plasmid vector, the effects of IL 4 secreted by the cells and γ ray irradiation on the secreting function of the cancer cells were detected. Results: The productive insertion of IL 4 gene was achieved. Gene transfection markedly enhanced the tumor killing effects of LAK cells activated by IL 4. After γ ray irradiation, the transfected cells continued to secrete IL 4 for about 3 to 4 weeks. Conclusion: Transfection of human IL 4cDNA can not change the in vitro growth characteristics of cancer cells, but it can significantly enhance antitumor immune response. γ ray irradiation might be a safe measure in the treatment of gastric cancer with IL 4 gene.
