摘要
目的:探索胃泌素(Gastrin)对胆管癌细胞增殖的调节作用。方法:应用细胞培养技术及3H-Tdr掺入法,研究了胃泌素-17(G-17)、亮氨酸胃泌素-17(LeuG-17)、五肽胃泌素(G-5)及胆囊收缩素(CCK-B/Gastrin)受体拮抗剂L-365,260(L-60)对培养的人胆管癌细胞QBC939(QB)、OCUCh-LM1(OC)、HuCC-T1(Hu)、OZ增殖的影响。结果:G-17、LeuG-17、G-5通过CCK-B/Gastrin受体显著的促进了QB、OC的增殖,而对Hu和OZ的增殖无明显影响。结论:部分胆管癌细胞有CCK-B/Gastrin受体表达,胃泌素可促进胆管癌细胞的增殖,为发展胆管癌的生物学治疗方式提供了新的资料。
Objective: To detect the regulative effects of gastrin on the proliferation of human cholangiocarcinoma cells. Methods: The effects of Gastrin 17 (G 17), Leu Gastrin 17 (LeuG 17), pentagastrin (G 5) and cholecystokinin (CCK) B/Gastrin receptor antagonist L365, 260 (L 60) on the proliferation of human cholangiocarcinoma cell lines QBC939 (QB), OCUCh LM1 (OC), HuCC T1 (Hu) and OZ were investigated with cell culture and 3HTdR incorporation. Results: G 17, LeuG 17 and G 5 significantly stimulated cell growth of QB and OC through receptor of CCK B/Gastrin, but the same result was not duplicated for Hu and OZ cell lines. Conclusion: Some human cholangiocarcinoma cell lines can express CCK B/Gastrin receptor, identifying that gastrin can accelerate the proliferation of human cholangiocarcinoma cells. This provides new theoretical basis for biological treatment of cholangiocarcinoma.
